Granulocyte-macrophage colony-stimulating factor, often abbreviated to GM-CSF, is a protein secreted by macrophages, T cells, mast cells, endothelial cells, and fibroblasts. GM-CSF contains N-linked and possibly O-linked oligosaccharides.
GM-CSF functions
GM-CSF stimulates stem cells to produce granulocytes (neutrophils, eosinophils, and basophils) and monocytes. GM-CSF is known as molgramostim or, when the protein is expressed in yeast cells, sargramostim (Leukine). GM-CSF is used as a medication to stimulate the production of white blood cells following chemotherapy. GM-CSF has also recently been evaluated in clinical trials for its potential as a vaccine adjuvant in HIV-infected patients. The preliminary results have been promising but GM-CSF is not presently FDA-approved for this purpose.

Molecular Weight: GM-CSF migrates as a broad band between 15 and 30 kDa on SDS-PAGE due to post-translation modifications, in particular glycosylation. This compares with unmodified GM-CSF that has a predicted molecular mass of 14.5 kDa. Purified GM-CSF consists of 5-50% carbohydrate by weight.
Production: rhGM-CSF is produced in E. coli and purified via sequential chromatography. Filtered prior to lyophilization through a 0.22 micron sterile filter.
Biological Activity: ED50 range = 0.02 to 0.1 ng/mL, determined by the dose dependent proliferation of human TF-1 cells. Optimal concentration for individual application should be determined by a dose response assay.
